Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Storage stability depends on water content, oxygen exposure, and temperature. Lyophilized solids are hygroscopic and can absorb water if exposed to humid air. Vials are usually sealed under vacuum or inert gas with rubber stoppers and aluminum crimps. Storage temperatures range from room temperature to refrigerated or frozen conditions depending on the material. Stability studies track potency, moisture, and physical form over time. Accelerated conditions can reveal sensitivity but may not predict long-term behavior.
Analytical methods for lyophilized materials include X-ray diffraction for crystallinity, differential scanning calorimetry for thermal transitions, and scanning electron microscopy for pore morphology. Moisture sorption analysis shows how the cake responds to humidity. These methods help distinguish amorphous from crystalline states and detect phase changes. Open questions remain about how pore structure changes during long-term storage and how best to predict collapse under varied conditions. Comparisons across studies are complicated by differences in formulation, cycle, and storage history.
Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.
| Property | Value | Notes |
|---|---|---|
| Appearance | Porous cake or plug | Uniform structure suggests the drying cycle preserved the matrix. |
| Reconstitution time | Usually under 2 minutes | Depends on cake porosity, diluent volume, and excipient composition. |
| Water content range | 0.5–3% w/w | Common specification range; exact limits are product-specific. |
| Headspace oxygen | <1% v/v | Inert gas backfill reduces oxidation of sensitive materials. |
| Storage temperature | 2–8 °C or controlled room temperature | Choice depends on accelerated and real-time stability results. |
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture uptake. Residual water content is measured because small changes can alter chemical stability and cake appearance. Storage temperature depends on the material; many biological products are kept at 2–8 °C, while some require −20 °C or colder. Exposure to ambient humidity during handling can cause the porous solid to absorb water and collapse. Container closures and stoppers are therefore selected for low moisture transmission and compatibility.
Quality control for lyophilized materials includes visual inspection of the cake, measurement of residual moisture, and tests for reconstitution time. An acceptable cake is typically uniform and may be slightly porous; shrinkage, meltback, or cracks can indicate process deviations. Analytical methods such as Karl Fischer titration, thermogravimetric analysis, and near-infrared spectroscopy quantify water content. Reconstitution time is recorded because a very slow or incomplete dissolution can signal collapse or aggregation. Stability studies compare samples stored under defined temperature and humidity conditions over months or years.
After lyophilization, the product is usually a porous cake or powder with a large internal surface area. This structure can absorb moisture quickly if exposed to humid air, so vials are sealed under vacuum or an inert gas. Moisture uptake may lower the glass transition temperature of the dried matrix and accelerate chemical or physical degradation. Storage conditions therefore depend on the formulation, container, and intended shelf life. Some products remain stable at room temperature, while others require refrigeration or freezing.
Quality control for lyophilized products includes appearance, cake structure, reconstitution time, pH, residual moisture, and potency. Residual moisture is a key attribute because excess water can reduce stability, while excessively low moisture may cause structural changes or aggregation in some systems. Stability studies compare real-time and accelerated conditions to estimate shelf life. Analytical methods must be validated for the specific matrix, container, and moisture range. Sterility and container integrity are also monitored for sterile products.
Handling practices aim to prevent moisture ingress and mechanical damage. Vials should remain stoppered and crimped until use, and reconstitution should follow the labeled diluent and volume. Shipping may involve temperature-controlled containers and desiccants, but direct contact between desiccant and product is avoided. Regulatory guidance expects documented storage conditions, excursion assessments, and stability commitments. Open questions remain about how best to predict long-term stability from short accelerated studies for every formulation class.
Quality control for lyophilized materials examines appearance, reconstitution time, residual moisture, and mechanical integrity. An acceptable cake is usually uniform and porous, though appearance alone does not prove stability. Karl Fischer titration is a common method for water content, while differential scanning calorimetry can reveal glass transition events. Stability studies track potency, aggregation, and moisture over time under defined temperature and humidity conditions. Specifications are product-specific and may include sterility and endotoxin tests for sterile preparations.
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
totipotency A state of cell potency in which a cell or nucleus fully retains the ability to differentiate into all of the cell types represented in the adult organism, or to give rise to all of these cell types upon transplantation into an appropriate cytoplasm (as in nuclear transfer). Such cells or nuclei are said to be totipotent. The zygote that serves as the progenitor cell for sexually reproducing multicellular organisms is the archetypal totipotent cell; almost all of the cells into which it ultimately differentiates are not totipotent, though some cells such as stem cells remain totipotent or pluripotent throughout the organism's life.
An ECG, which is a recording of the heart's electrical activity, may confirm an ST elevation MI (STEMI), if ST elevation is present. Commonly used blood tests include troponin and less often creatine kinase MB. Treatment of an MI is time-critical. Aspirin is an appropriate immediate treatment for a suspected MI. Nitroglycerin or opioids may be used to help with chest pain; however, they do not improve overall outcomes. Supplemental oxygen is recommended in those with low oxygen levels or shortness of breath. In a STEMI, treatments attempt to restore blood flow to the heart and include percutaneous coronary intervention (PCI), where the arteries are pushed open and may be stented, or thrombolysis, where the blockage is removed using medications. People who have a non-ST elevation myocardial infarction (NSTEMI) are often managed with the blood thinner heparin, with the additional use of PCI in those at high risk. In people with blockages of multiple coronary arteries and diabetes, coronary artery bypass surgery (CABG) may be recommended rather than angioplasty. After an MI, lifestyle modifications, along with long-term treatment with aspirin, beta blockers and statins, are typically recommended. Worldwide, about 15.9 million myocardial infarctions occurred in 2015. More than 3 million people had an ST elevation MI, and more than 4 million had an NSTEMI. STEMIs occur about twice as often in men as women. About one million people have an MI each year in the United States. In the developed world, the risk of death in those who have had a STEMI is about 10%.
In early 1987, he persuaded 11 Doobie alumni to join him for a concert to benefit veterans' causes. Answering the call were Tom Johnston, Pat Simmons, Jeff Baxter, John McFee, John Hartman, Michael Hossack, Chet McCracken, Michael McDonald, Cornelius Bumpus, Bobby LaKind and Tiran Porter, plus their long-time record producer and good friend, Ted Templeman. There were no surplus bass players as Weeks had other commitments. They soon discovered that tickets were in great demand, so the concert quickly evolved into a 12-city tour that began on May 21, 1987, in San Diego. The third concert, held at the Hollywood Bowl, was reportedly the venue's fastest sell-out since the Beatles had played there just over 20 years earlier. The band performed selections from every album using a wide variety of instrumentation that they could not have previously duplicated onstage without the expanded lineup. Baxter and McFee played pedal steel and violin, respectively, during "Black Water" and "Steamer Lane Breakdown". "Without You" featured four drummers and four lead guitarists. Producer Ted Templeman played percussion and LaKind sometimes played Knudsen's drum set while Knudsen went to the front of the stage to join the chorus. Templeman also played the drum set on "What a Fool Believes", as he did on the original hit record. The tour culminated (sans McDonald, LaKind, McFee and Knudsen) at the Glasnost-inspired July 4 "Peace Concert" in Moscow, with Bonnie Raitt, James Taylor and Santana sharing the bill.
In intracellular chemistry, sulfur operates as a carrier of reducing hydrogen and its electrons for cellular repair of oxidation. Reduced glutathione, a sulfur-containing tripeptide, is a reducing agent through its sulfhydryl (–SH) moiety derived from cysteine. Sulfur atoms may be catenated on the sulfhydryl groups of cysteine or glutathione to form a hydropersulfide (–SSH), which act as signaling molecules and donors of sulfur atoms in the biosynthesis of iron-sulfur clusters and some sulfur containing metabolites. Polysulfides and elemental sulfur are also known to exist in both prokaryotes and eukaryotes. Methanogenesis, the route to most of the world's methane, is a multistep biochemical transformation of carbon dioxide. This conversion requires several organosulfur cofactors. These include coenzyme M, CH3SCH2CH2SO−3, the immediate precursor to methane.

== Detection in alcoholic beverages == The concerns raised by the toxicological aspects of EC together with the low concentration levels (μg/L) found in wines, as well as the occurrence of interferences on detection, has motivated several researchers to develop new methods to determine it in wines. Several extraction and chromatographic techniques have been used, including continuous liquid–liquid extraction (LLE) with Soxhlet apparatus, derivatization with 9-xanthydrol followed by high-performance liquid chromatography (HPLC) with fluorescence detection and even LLE after derivatization, followed by gas chromatography coupled with mass spectrometry detection (GC–MS). On the other hand, the reference method set by the International Organization of Vine and Wine (OIV) uses solid phase extraction (SPE) preceding GC–MS quantification. Other methods also make use of SPE, but use gas chromatography with mass spectrometry (MDGC/MS) and liquid chromatography with tandem mass spectrometry (LC–MS/MS) for detection. Most of the methodologies found in the literature to quantify EC use gas chromatography, using LLE and SPE as extraction techniques. Nevertheless, several efforts have also been done to develop new methodologies to determine EC without using long procedures and hard-working analyses, combining precision to high sensitivity.
In the Histidine variant, the enzyme is much more effective at the aforementioned conversion. The enzyme responsible for the conversion of acetaldehyde to acetate, however, remains unaffected, which leads to differential rates of substrate catalysis and causes a buildup of toxic acetaldehyde, causing cell damage. This provides some protection against excessive alcohol consumption and alcohol dependence (alcoholism). Various haplotypes arising from this mutation are more concentrated in regions near Eastern China, a region also known for its low alcohol tolerance and dependence. A study was conducted in order to find a correlation between allelic distribution and alcoholism, and the results suggest that the allelic distribution arose along with rice cultivation in the region between 12,000 and 6,000 years ago. In regions where rice was cultivated, rice was also fermented into ethanol. This led to speculation that increased alcohol availability led to alcoholism and abuse, resulting in lower reproductive fitness. Those with the variant allele have little tolerance for alcohol, thus lowering chance of dependence and abuse. The hypothesis posits that those individuals with the Histidine variant enzyme were sensitive enough to the effects of alcohol that differential reproductive success arose and the corresponding alleles were passed through the generations. Classical Darwinian evolution would act to select against the detrimental form of the enzyme (Arg variant) because of the lowered reproductive success of individuals carrying the allele.
=== Imaging === The SBP-Tag will also bind to streptavidin or streptavidin reagents in solution. Applications of these engineered associations include the visualization of specific proteins within living cells, monitoring of the kinetics of the translation of individual proteins in an in vitro translation system, control of the integration of a multi-spanning membrane protein into the endoplasmic reticulum by fusing the SBP-Tag to the N-terminal translocation sequence and then halting integration with streptavidin and restarting integration with biotin. Fluorescent streptavidin reagents (e.g. streptavidin-HRP) can be used to visualize the SBP-tag by immunoblotting of SDS-PAGE. Additionally, antibodies to the SBP-tag are available commercially.

As part of decolonization, France established the French Union in 1946 and reorganized Indochina in 1949 as a confederation of associated states within it. In June 1949, they united French Cochinchina with the protectorates of Annam and Tonkin to form the State of Vietnam within the French Union, and installed former Emperor Bảo Đại as head of state. The Chinese Communist victory of 1949 changed the nature of the Indochina War. In 1950, the newly established People's Republic of China and the Soviet Union recognized the DRV while the United States recognized the State of Vietnam. The conflict largely resembled a conventional war between two armies equipped with modern weapons, although guerrilla warfare continued to occur in many areas. The United States provided assistance to France, while China assisted the Viet Minh. French Union forces included colonial troops from the empire – North Africans, Sub-Saharan Africans, Laotian, Cambodian and Vietnamese ethnic minorities – and professional French troops, European volunteers, and units of the Foreign Legion. The use of French metropolitan recruits was forbidden by the government to prevent the war from becoming more unpopular at home. It was called the "dirty war" (la sale guerre) by French leftists. In December 1950, France officially established an army for the State of Vietnam. In September 1951, the US began providing direct economic aid to the State of Vietnam. Segments of the population supported neither the communist Viet Minh nor the French colonialists, and were labeled attentistes.

=== Unknown blood type === Because blood type O negative is compatible with anyone, it is often overused and in short supply. According to the Association for the Advancement of Blood and Biotherapies, the use of this blood should be restricted to persons with O negative blood, as nothing else is compatible with them, and women who might be pregnant and for whom it would be impossible to do blood group testing before giving them emergency treatment. Whenever possible, the AABB recommends that O negative blood be conserved by using blood type testing to identify a less scarce alternative.

Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
It usually appears as a uniform porous plug or cake that fills the container without excessive shrinkage. Color should match the specification, and there should be no meltback or visible foreign matter. Minor cracking may be acceptable if the product still meets moisture and potency limits.
Water content is a key stability parameter because excess water can promote hydrolysis, aggregation, or cake collapse. It also affects reconstitution and product weight. Each product has a target range, and methods such as Karl Fischer titration are used to verify it.
Karl Fischer titration is a common method, using coulometric or volumetric detection. Thermogravimetric analysis can also measure weight loss on heating. Results depend on sample handling because the dried solid can absorb moisture quickly.
