Reconstitution involves adding a suitable diluent, often sterile water or a buffer, to the dried cake. Gentle swirling or inversion helps dissolve the material without creating excessive foam. The time required for complete dissolution can range from seconds to several minutes and depends on the cake structure and the diluent. Improper reconstitution, such as vigorous shaking or using the wrong diluent, can cause protein aggregation or loss of activity. After reconstitution, the product may have a limited shelf life and should be used according to its labeling.
Lyophilized products are typically hygroscopic and require protection from moisture during storage. Manufacturers seal them in glass vials, often under vacuum or an inert gas such as nitrogen. The container closure system, including the stopper and crimp seal, must prevent water vapor ingress. Storage temperature varies from controlled room temperature to refrigerated or frozen conditions, depending on the formulation. Humidity-controlled environments are essential because even brief exposure to ambient air can degrade the product.
A freeze-dryer consists of a vacuum chamber, temperature-controlled shelves, a condenser, and a vacuum pump. Vials, ampoules, or bulk trays hold the product during the cycle. The condenser traps water vapor as ice at a temperature lower than the product. Cycle development balances shelf temperature, chamber pressure, and time. Scale-up can be difficult because heat and mass transfer change with equipment size, so process analytical tools and conservative validation are often used.
Lyophilization is a dehydration technique in which a product is frozen and the solvent is removed under reduced pressure. The low pressure allows ice to sublimate directly into vapor without passing through a bulk liquid phase. This differs from conventional drying, where heat drives evaporation and can damage heat-sensitive structures. The process is used for biological materials, pharmaceutical formulations, and some foods. Its main advantage is preservation of porous structure and rapid reconstitution.
| Property | Value | Notes |
|---|---|---|
| Appearance | White to off-white porous cake | Color depends on formulation. |
| Typical storage temperature | 2–8 °C | Refrigerated for many biologics. |
| Residual moisture | <1% to 3% | Low moisture improves stability. |
| Container | Sealed glass vial | Often with rubber stopper and aluminum crimp. |
| Reconstitution time | Seconds to minutes | Varies with cake density and diluent. |
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Secondary drying removes bound water that remains after ice sublimation. Shelf temperature is raised gradually while pressure remains low, reducing water content to a target range. Over-drying can cause brittleness or electrostatic issues, while under-drying affects stability. The endpoint is often judged by pressure rise tests, temperature measurements, or water content analysis. Scale-up depends on matching heat and mass transfer across equipment sizes. Small changes in shelf temperature or pressure can alter cycle length substantially.
Lyophilization, or freeze-drying, removes water from a material by freezing it and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intervening liquid state. It is used for heat-sensitive materials that would degrade in conventional drying. The three stages are freezing, primary drying, and secondary drying, each with distinct temperature and pressure requirements. In practice, cycle design balances these variables.
Freezing determines ice crystal structure and pore size, which affect drying speed and product uniformity. Rapid freezing creates small crystals, while slow freezing creates larger crystals and often faster sublimation. During primary drying, chamber pressure is held below the vapor pressure of ice, and shelf temperature supplies heat for sublimation. The ice front recedes, leaving a porous matrix. Thermal limits such as collapse and eutectic temperatures set safe boundaries for formulation. These limits vary with solute composition and concentration.
Handling practices aim to prevent moisture ingress and mechanical damage. Vials should remain stoppered and crimped until use, and reconstitution should follow the labeled diluent and volume. Shipping may involve temperature-controlled containers and desiccants, but direct contact between desiccant and product is avoided. Regulatory guidance expects documented storage conditions, excursion assessments, and stability commitments. Open questions remain about how best to predict long-term stability from short accelerated studies for every formulation class.
After lyophilization, the product is usually a porous cake or powder with a large internal surface area. This structure can absorb moisture quickly if exposed to humid air, so vials are sealed under vacuum or an inert gas. Moisture uptake may lower the glass transition temperature of the dried matrix and accelerate chemical or physical degradation. Storage conditions therefore depend on the formulation, container, and intended shelf life. Some products remain stable at room temperature, while others require refrigeration or freezing.
Quality control for lyophilized products includes appearance, cake structure, reconstitution time, pH, residual moisture, and potency. Residual moisture is a key attribute because excess water can reduce stability, while excessively low moisture may cause structural changes or aggregation in some systems. Stability studies compare real-time and accelerated conditions to estimate shelf life. Analytical methods must be validated for the specific matrix, container, and moisture range. Sterility and container integrity are also monitored for sterile products.
In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
A typical cycle begins with freezing, which fixes the material into a solid and determines ice crystal size. Primary drying then raises heat under vacuum so ice sublimes, often near or below the collapse temperature of the formulation. Secondary drying removes bound water that remains after ice is gone, usually by gently warming the product. Each stage balances heat input against pressure to avoid melting or structural damage. Temperature probes and pressure sensors guide the transition between stages.
=== 2016–present: Departure from Valve === Laidlaw announced his departure from Valve in January 2016. He said the primary reason for his departure was his age, and that he planned to return to writing stories. He felt he was becoming a "negative force" at Valve and hampering the creative process, saying: "I think at some point you need to let the people who are the fans and the creators who've come in because of what they learned from you maybe, and let them have that. We didn't need me going, 'Well, the G-Man wouldn't do that in my day.'" Laidlaw also tired of the FPS genre and of solving storytelling problems in a Half-Life-style narrative. He said he had "always hoped that we'd stumble into a more expansive vocabulary or grammar for storytelling within the FPS medium, one that would let you do more than shoot or push buttons, or push crates". On August 25, 2017, Laidlaw published a short story, "Epistle 3", describing it as "a snapshot of a dream I had many years ago". Journalists interpreted it as a summary of what could have been the plot for Half-Life 2: Episode Three. Laidlaw denied this, saying "all the real story development can only happen in the crucible of developing the game". In 2023, Laidlaw said he regretted publishing the story. He said he had been "deranged" and "completely out of touch" at the time, and that it had created problems for his former colleagues at Valve. In 2020, Valve released the VR game Half-Life: Alyx.
Desomorphine (or in some formulations known as Krokodil) is a semi-synthetic opioid commercialized by Roche, with powerful, fast-acting effects, such as sedation and analgesia. It was first discovered and patented in Germany by a German team working for Knoll in 1920 but was not generally recognized. It was later synthesized in 1932 by American chemist Lyndon Frederick Small. Small also successfully patented it in 1934 in the United States. Desomorphine was used in Germany, Austria, and Switzerland under the brand name Permonid and was described as having a fast onset and a short duration of action, with relatively little nausea compared to equivalent doses of morphine. Dose for dose it is roughly ten times more potent than morphine, with 1 mg desomorphine being equivalent 10 mg morphine, via the intravenous (IV) or intramuscular (IM) routes. Desomorphine is a morphine analogue where the 6-hydroxyl group and the 7,8 double bond have been reduced. The traditional synthesis of desomorphine starts from α-chlorocodide, which is itself obtained by treating codeine with thionyl chloride. By catalytic reduction, α-chlorocodide gives dihydrodesoxycodeine, which yields desomorphine on demethylation. A desomorphine product, usually based on codeine, has been developed as a recreational drug. The product in question is typically a highly impure variant of desomorphine. The scaly sores and necrosis that develop around the injection site has prompted the name krokodil (Russian for crocodile).
=== Analysis === Northeastern University School of Law professor Alexandra Roberts argued that Twitter's claim that certain individuals paid for Twitter Blue may violate state and federal false representation laws, such as the Lanham Act, popularized through a retweet by dril, but reserved that Twitter was not making a direct advertisement. Solicitor Simon McGarr mentioned that Twitter's insistence on the blue checkmark for some users, such as dril, may violate the General Data Protection Regulation (GDPR) in the European Union, as verifying accounts requires a phone number. McGarr also noted Tolley v. Fry, a 1931 court case filed against chocolate bar manufacturer J. S. Fry & Sons by golfer Cyril Tolley alleging that the advertising of chocolate bars with a caricature of Tolley was defamatory, or the similar case Eddie Irvine v. Talksport between racing driver Eddie Irvine and sports radio station Talksport. Other scholars have cited the Federal Trade Commission Act of 1914, while some have pointed to the disdain for blue checkmarks and the potential effects of the checkmark on celebrities' reputations. The Federal Trade Commission (FTC) has not commented on Twitter Blue, although an anonymous former FTC official called it "deceptive". Following the initial implementation of paid verification, the Twitter account for the anti-vaccine propaganda film Died Suddenly (2022) became verified. The account has been used to spread medical misinformation, including of COVID-19 vaccines.

The first point to note is that, when pKa is positive, the standard free energy change for the dissociation reaction is also positive. Second, some reactions are exothermic and some are endothermic, but, when ΔH⊖ is negative TΔS⊖ is the dominant factor, which determines that ΔG⊖ is positive. Last, the entropy contribution is always unfavourable (ΔS⊖ < 0) in these reactions. Ions in aqueous solution tend to orient the surrounding water molecules, which orders the solution and decreases the entropy. The contribution of an ion to the entropy is the partial molar entropy which is often negative, especially for small or highly charged ions. The ionization of a neutral acid involves formation of two ions so that the entropy decreases (ΔS⊖ < 0). On the second ionization of the same acid, there are now three ions and the anion has a charge, so the entropy again decreases. Note that the standard free energy change for the reaction is for the changes from the reactants in their standard states to the products in their standard states. The free energy change at equilibrium is zero since the chemical potentials of reactants and products are equal at equilibrium.
=== Clinical trials === Human trials began on January 26, 2021, with 60 volunteers between the ages of 18 and 65 in Toronto. Of these, 15 would receive a placebo and three groups of 15 would receive different doses of the vaccine. The volunteers will be monitored for 13 months. The company said that enough data would be available in May which could result in a Phase 2 clinical testing beginning soon after that, pending regulatory approval. If the results of a subsequent larger human trial are positive, the vaccine could enter a commercialization phase in 2022. The Phase 1 clinical trial lead was Piyush Patel. At the 29 April meeting with the House of Commons, Sorenson estimated that PTX-COVID19-B could be approved by Health Canada by "January or February 2022".
McMillan responded, "at this point, Marvel's brand is such that I'm not sure it can offer up something like [the trailer] without it seeming like a crushing disappointment ... part of Marvel's brand is that it doesn't offer the kind of run-of-the-mill superhero movie that you're talking about, that it's ... at least different enough to tweak and play with the genre somehow ... The fact that there's such upset about this trailer being ... well, okay ... suggests to me that the audience is expecting something to knock their socks off." Doran concluded, "That does seem to be the point here—the expectations fans now have for everything Marvel Studios ... [and] Marvel is going to eventually falter." After seeing the portrayal of Yellowjacket in Ant-Man, the antagonist of the film, McMillan noted,
Primary structure Elzinga and co-workers first determined the complete peptide sequence for this type of actin in 1973, with later work by the same author adding further detail to the model. It contains 374 amino acid residues. Its N-terminus is highly acidic and starts with an acetyled aspartate in its amino group, while its C-terminus is alkaline and is formed by a phenylalanine preceded by a cysteine, which has a degree of functional importance. Both extremes are in close proximity within the I-subdomain. An anomalous Nτ-methylhistidine is located at position 73.

Hypoprolactinemia, or serum prolactin deficiency, is associated with ovarian dysfunction in women, and arteriogenic erectile dysfunction, premature ejaculation, oligozoospermia, asthenospermia, hypofunction of seminal vesicles and hypoandrogenism in men. In one study, normal sperm characteristics were restored when prolactin levels were raised to normal values in hypoprolactinemic men. Hypoprolactinemia can result from hypopituitarism, excessive dopaminergic action in the tuberoinfundibular pathway and ingestion of D2 receptor agonists such as bromocriptine. Dextroamphetamine has been shown to reduce serum prolactin, presumably due to its dopaminergic effects, indirectly increasing activation of dopamine D2 receptors with data showing intravenous dextroamphetamine at doses between 7.5 mg to 15 mg reducing serum prolactin by 27-32% and 30-37%, respectively, while an oral dose of 20 mg led to a sustained 40% reduction of serum prolactin in postpartum women.
Active hexose correlated compound (AHCC) is an alpha-glucan rich nutritional supplement produced from shiitake (Lentinula edodes). The product is a subject of research as a potential anti-cancer agent. AHCC is a popular alternative medicine in Japan. AHCC is a registered trademark of and manufactured by Amino Up Co., Ltd. in Sapporo City, Hokkaido, Japan. AHCC was developed by Amino Up Co., LTD. and Toshihiko Okamoto (School of Pharmaceutical Sciences, University of Tokyo) in 1989. Polysaccharides form a large part of the composition of AHCC. These include beta-glucan (β-glucan) and partially acylated α-glucan. Partially acylated α-glucan, produced by the patented long term culturing process, is unique to AHCC. Approximately 20% of the make up of AHCC is α-glucans. Glucans are saccharides, of which some are known to have immune stimulating effects.
=== Emergence of the Israelite culture === Efforts to confirm the biblical ethnogenesis of Israel through archaeology have largely been abandoned as unfruitful. Many scholars see the traditional narratives as national myths with little historical value, but some posit that a small group of exiled Egyptians contributed to the Exodus narrative. William G. Dever cautiously identifies this group with the Tribe of Joseph, while Richard Elliott Friedman identifies it with the Tribe of Levi. Josephus quoting Manetho identifies them with the Hyksos. Other scholars believe that the Exodus narrative was a "collective memory" of several events from the Bronze Age.
By the end of March 2021 RDIF anticipates 33 million doses will have been manufactured in Russia, less than 5% of which will have been exported. An agreement for the production of over 100 million doses of vaccine in India was made with Dr. Reddy's Laboratories, which on 11 January 2021 submitted mid-stage trial data to the Indian regulator and recommended moving onto late-stage trials. The RDIF announced plans to sell 100 million doses to India, 35 million to Uzbekistan, and 32 million to Mexico, as well as 25 million each to Nepal and Egypt. In India, the first dose of Sputnik V vaccine was administered on 14 May 2021 at Hyderabad. Argentina became the first Latin American country to produce it. Large-scale production started in June 2021. As of 31 December 2021 277 million doses were manufactured, mostly (265 million) in Russia. On 28 February 2022, as a result of the 2022 Russian invasion of Ukraine, the United States placed RDIF and its chief executive on its list of sanctioned Russian entities and people. The European Union, Ukraine, United Kingdom and Australia followed later in February and in March. This significantly reduces vaccine's future commercial prospects.

Lyophilized products should be stored in airtight containers, protected from moisture and light, at the temperature specified by the manufacturer. Many require refrigeration at 2–8 °C, while some need frozen storage. Always check the product label for specific conditions.
Moisture can cause the porous cake to collapse, increase molecular mobility, and accelerate chemical degradation. It may also promote microbial growth if the product lacks preservatives. Proper sealing and handling are essential to maintain stability.
Some formulations contain labile biological molecules that degrade even in the dry state at higher temperatures. Others have a low glass transition temperature, meaning the cake can soften or collapse at room temperature. Cold storage reduces molecular motion and slows degradation.
Yes, the terms are generally interchangeable. Lyophilization is more common in pharmaceutical and laboratory contexts, while freeze-drying appears widely in food science and general writing. Both describe removal of solvent by sublimation under vacuum after freezing.
