In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
A typical cycle begins with freezing, which fixes the material into a solid and determines ice crystal size. Primary drying then raises heat under vacuum so ice sublimes, often near or below the collapse temperature of the formulation. Secondary drying removes bound water that remains after ice is gone, usually by gently warming the product. Each stage balances heat input against pressure to avoid melting or structural damage. Temperature probes and pressure sensors guide the transition between stages.
Quality control for lyophilized products includes appearance, cake structure, reconstitution time, pH, residual moisture, and potency. Residual moisture is a key attribute because excess water can reduce stability, while excessively low moisture may cause structural changes or aggregation in some systems. Stability studies compare real-time and accelerated conditions to estimate shelf life. Analytical methods must be validated for the specific matrix, container, and moisture range. Sterility and container integrity are also monitored for sterile products.
Handling practices aim to prevent moisture ingress and mechanical damage. Vials should remain stoppered and crimped until use, and reconstitution should follow the labeled diluent and volume. Shipping may involve temperature-controlled containers and desiccants, but direct contact between desiccant and product is avoided. Regulatory guidance expects documented storage conditions, excursion assessments, and stability commitments. Open questions remain about how best to predict long-term stability from short accelerated studies for every formulation class.
| Property | Value | Notes |
|---|---|---|
| Common synonym | Freeze-drying | Same dehydration operation |
| Typical vacuum | 10-100 Pa | Pressure during primary drying |
| Primary drying temperature | -40 to -10 °C | Below collapse temperature for many formulations |
| Cycle duration | 12-72 hours | Varies with load, container, and formulation |
| Key phase change | Sublimation | Solid ice to water vapor |
Secondary drying removes bound water that remains after ice sublimation. Shelf temperature is raised gradually while pressure remains low, reducing water content to a target range. Over-drying can cause brittleness or electrostatic issues, while under-drying affects stability. The endpoint is often judged by pressure rise tests, temperature measurements, or water content analysis. Scale-up depends on matching heat and mass transfer across equipment sizes. Small changes in shelf temperature or pressure can alter cycle length substantially.
Lyophilization, or freeze-drying, removes water from a material by freezing it and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intervening liquid state. It is used for heat-sensitive materials that would degrade in conventional drying. The three stages are freezing, primary drying, and secondary drying, each with distinct temperature and pressure requirements. In practice, cycle design balances these variables.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
A large systematic review concluded that circulating IGFBP-3 levels showed a modest association with increased risk for a number of cancers, but the results vary among sites. IGFBP-3 protein levels decrease during the progression of prostate cancer from benign to metastatic disease although production of the protein does not cease completely. IGFBP-3 is still made (at a lower level) by prostate cancer cells and secreted into the surrounding environment. However, instead of the full length, functional protein, IGFBP-3 is found to be cleaved. This decreases the affinity of IGF binding to IGFBP-3, making the growth factors more likely to bind the IGF1R and promote cell survival.
Changes in actin's folding occur in nemaline myopathy as well as changes in its aggregation and there are also changes in the expression of other associated proteins. In some variants where intranuclear bodies are found the changes in the folding masks the nucleus's protein exportation signal so that the accumulation of actin's mutated form occurs in the cell nucleus. On the other hand, it appears that mutations to ACTA1 that give rise to a CFTDM have a greater effect on sarcomeric function than on its structure. Recent investigations have tried to understand this apparent paradox, which suggests there is no clear correlation between the number of rods and muscular weakness. It appears that some mutations are able to induce a greater apoptosis rate in type II muscular fibres.
The U.S. has worked with Guatemalan authorities to clamp down on South American cocaine routes, many of which use Guatemala as a landing zone. In October 2013, the US supplied six twin-engine "Super Huey" helicopters to Guatemala in an effort to halt illegal air traffic.
Straight-chain alkanes are sometimes indicated by the prefix n- (for "normal") where a non-linear isomer exists. Although this is not strictly necessary and is not part of the IUPAC naming system, the usage is still common in cases where one wishes to emphasize or distinguish between the straight-chain and branched-chain isomers, e.g., "n-butane" rather than simply "butane" to differentiate it from isobutane. Alternative names for this group used in the petroleum industry are linear paraffins or n-paraffins. The first eight members of the series (in terms of number of carbon atoms) are named as follows:

An interesting alternative synthesis of this substance starts by sequential reaction of N-benzylaniline with phosgene, and then with sodium azide to produce the corresponding carbonyl azide. On heating, nitrogen is evolved and a separatable mixture of nitrene insertion product and the desired ketoindazole # results. The latter reaction appears to be a Curtius rearrangement type product to produce an N-isocyanate #, which then cyclizes. Alkylation of the enol with sodium methoxide and 3-dimethylaminopropyl chloride gives benzydamine. Alternatively, use of chloroacetamide in the alkylation step followed by acid hydrolysis produces bendazac instead.
=== Nutritional content === At birth, the composition of breast milk corresponds in quality to the specific needs of the infant. The pattern of intended nutrient content in breast milk is relatively consistent. Breastmilk is made from nutrients in the mother's bloodstream and bodily stores. It has an optimal balance of fat, sugar, water, and protein that is needed for a baby's age-appropriate growth and development. That being said, a variety of factors can influence the nutritional makeup of breastmilk, including gestational age, age of infant, maternal age, maternal smoking, and nutritional needs of the infant. The first type of milk produced is called colostrum. The volume of colostrum produced during each feeding is appropriate for the size of the newborn's stomach and is sufficient, calorically, for feeding a newborn during the first few days of life. Produced during pregnancy and the first days after childbirth, colostrum is rich in protein and Vitamins A, B12 and K, which support infants' growth, brain development, vision, immune systems, red blood cells, and clotting cascade. The breast milk also has long-chain polyunsaturated fatty acids which help with normal retinal and neural development. The caloric content of colostrum is about 54 Calories/100mL. The second type of milk is transitional milk, which is produced during the transition from colostrum to mature breast milk. As the breast milk matures over several weeks, the protein content decreases on average.

== Wife of the heir to Austrian throne == At this time, Archduke Charles was in his twenties and did not expect to become emperor for some time, especially while Franz Ferdinand remained in good health. This changed on 28 June 1914 when the heir and his wife Sophie were assassinated in Sarajevo by Bosnian Serb nationalists. Charles and Zita received the news by telegram that day. She said of her husband, "Though it was a beautiful day, I saw his face go white in the sun." In the war that followed, Charles was promoted to General in the Austro-Hungarian army, taking command of the 20th Corps for an offensive in Tyrol. The war was personally difficult for Zita, as several of her brothers fought on opposing sides in the conflict (Prince Felix and Prince René had joined the Austrian army, while Prince Sixtus and Prince Xavier lived in France before the war and enlisted in the Belgian army.) Also her country of birth, Italy, joined the war against Austria in 1915, and so rumours of the 'Italian' Zita began to be muttered. Even as late as 1917, the German ambassador in Vienna, Count Botho von Wedel-Jarlsberg, would write to Berlin saying "The Empress is descended from an Italian princely house... People do not entirely trust the Italian and her brood of relatives." At Franz Joseph's request, Zita and her children left their residence at Schloss Hetzendorf and moved into a suite of rooms at Schönbrunn Palace. Here, Zita spent many hours with the old Emperor on both formal and informal occasions, where Franz Joseph confided in her his fears for the future.
Since iron is primarily required for hemoglobin, iron deficiency anemia is the primary clinical manifestation of iron deficiency. Iron-deficient people will suffer or die from organ damage well before their cells run out of the iron needed for intracellular processes like electron transport. Macrophages of the reticuloendothelial system store iron as part of the process of breaking down and processing hemoglobin from engulfed red blood cells. Iron is also stored as a pigment called hemosiderin, which is an ill-defined deposit of protein and iron, created by macrophages where excess iron is present, either locally or systemically, e.g., among people with iron overload due to frequent blood cell destruction and the necessary transfusions their condition calls for. If systemic iron overload is corrected, over time the hemosiderin is slowly resorbed by the macrophages.

== Non-autotrophic pathways == Although no heterotrophs use carbon dioxide in biosynthesis, some carbon dioxide is incorporated in their metabolism. Notably pyruvate carboxylase consumes carbon dioxide (as bicarbonate ions) as part of gluconeogenesis, and carbon dioxide is consumed in various anaplerotic reactions. 6-phosphogluconate dehydrogenase catalyzes the reductive carboxylation of ribulose 5-phosphate to 6-phosphogluconate in E. coli under elevated CO2 concentrations.
==== Cancer health claims ==== The U.S. Food and Drug Administration initiated a process of reviewing and approving food and dietary supplement health claims in 1993. Reviews of petitions results in proposed claims being rejected or approved. If approved, specific wording is allowed on package labels. In 1999, a second process for claims review was created. If there is not a scientific consensus on the totality of the evidence, a Qualified Health Claim (QHC) may be established. The FDA does not "approve" qualified health claim petitions. Instead, it issues a Letter of Enforcement Discretion that includes very specific claim language and the restrictions on using that wording. The first QHCs relevant to vitamin E were issued in 2003: "Some scientific evidence suggests that consumption of antioxidant vitamins may reduce the risk of certain forms of cancer." In 2009, the claims became more specific, allowing that vitamin E might reduce the risk of renal, bladder and colorectal cancers, but with required mention that the evidence was deemed weak and the claimed benefits highly unlikely. A petition to add brain, cervical, gastric and lung cancers was rejected. A further revision, May 2012, allowed that vitamin E may reduce risk of renal, bladder and colorectal cancers, with a more concise qualifier sentence added: "FDA has concluded that there is very little scientific evidence for this claim." Any company product label making the cancer claims has to include a qualifier sentence.

== WADA proscription == In 2015, Radwa Arafa Abd Elsalam, an athlete from Egypt, underwent a doping control where ractopamine was identified in the sample. Elsalam was sanctioned by the Egyptian Anti-Doping Organisation (EGY-NADO) with a two year ineligibility period for non-intentional presence of a prohibited substance, in violation of Article 2.1 of the EGY-NADO rules. While not mentioned by name in the 2015 Prohibited List, ractopamine was determined to — much like clenbuterol, another β2-adrenergic agonist — constitute an Other Anabolic Agent prohibited under S1.2. Elsalam appealed the decision, claiming that ractopamine is used in the meat industry overseas, so she must have inadvertently consumed food contaminated with ractopamine. EGY-NADO accepted her appeal, reduced the sanction to six months, and provided the World Anti-Doping Agency (WADA) with records pertaining to the case. WADA filed an appeal with the Court of Arbitration for Sport (CAS) against EGY-NADO and Elsalam with respect to the appealed decision. WADA asserted ractopamine was "undisputed[ly]" an Other Anabolic Agent, that "[i]t is not sufficient for an athlete merely to make protestations of innocence and suggest that the prohibited substance must have entered her body in inadvertently", and that "her explanations are nothing more than mere speculation".
In 2009, in an article for The Atlantic entitled "The Revenge of Karl Marx", Hitchens frames the late-2000s recession in terms of Marx's economic analysis and notes how much Marx admired the capitalist system that he called for the end of, but says that Marx ultimately failed to grasp how revolutionary capitalist innovation was. Hitchens was an admirer of Che Guevara, yet in an essay written in 1997, he distanced himself from Che, and referred to the mythos surrounding him as a "cult". In 2004 he re-emphasized his positive view of Che, commenting that "[Che's] death meant a lot to me and countless like me at the time. He was a role model, albeit an impossible one for us bourgeois romantics insofar as he went and did what revolutionaries were meant to do – fought and died for his beliefs." He continued to regard Leon Trotsky and Vladimir Lenin as great men, and the October Revolution as a necessary event in the modernisation of Russia. In 2005, Hitchens praised Lenin's creation of "secular Russia" and his attacks on the Russian Orthodox Church, describing the church's power as "absolute warren of backwardness and evil and superstition". In God is not Great, Hitchens called Marxism his "own secular faith" that "has been shaken and discarded, not without pain." He referred to his period of Marxist faith as "when I was a Marxist." According to Andrew Sullivan, his last words were "Capitalism, downfall."

Primary drying removes ice by sublimation under vacuum. Secondary drying removes water that is bound to the material, often by warming the product after most ice has left. Both stages occur below temperatures that would cause unwanted melting.
Sublimation requires the solvent to remain solid so vapor leaves without passing through a liquid phase. If the product melts, the porous structure can collapse and drying becomes uneven. Maintaining frozen conditions preserves the intended physical form.
No, freeze-drying is a dehydration method, not a sterilization step. It can reduce water activity and limit microbial growth during storage, but it does not reliably kill microbes or remove endotoxins. Sterility must come from separate validated processes.
Collapse occurs when the product temperature rises above its collapse or eutectic temperature during drying. The frozen matrix loses structure, producing a shrunken or melted appearance. This can slow reconstitution and may affect stability.
