reconstitution raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2025-07-25 and is reviewed periodically as new material appears.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability of a lyophilized product depends on its glass transition temperature, the temperature at which the amorphous cake transitions from a glassy to a rubbery state. Storage below this temperature minimizes molecular mobility and slows chemical degradation. If the storage temperature exceeds the glass transition temperature, the cake may collapse, shrink, or become sticky. Accelerated stability studies at elevated temperatures and humidity help predict shelf life, but they do not always reflect real-time behavior. Residual moisture content also plays a critical role in long-term stability.
Reconstitution involves adding a suitable diluent, often sterile water or a buffer, to the dried cake. Gentle swirling or inversion helps dissolve the material without creating excessive foam. The time required for complete dissolution can range from seconds to several minutes and depends on the cake structure and the diluent. Improper reconstitution, such as vigorous shaking or using the wrong diluent, can cause protein aggregation or loss of activity. After reconstitution, the product may have a limited shelf life and should be used according to its labeling.
Lyophilized products are typically hygroscopic and require protection from moisture during storage. Manufacturers seal them in glass vials, often under vacuum or an inert gas such as nitrogen. The container closure system, including the stopper and crimp seal, must prevent water vapor ingress. Storage temperature varies from controlled room temperature to refrigerated or frozen conditions, depending on the formulation. Humidity-controlled environments are essential because even brief exposure to ambient air can degrade the product.
| Property | Value | Notes |
|---|---|---|
| Residual moisture | 0.5-3% w/w | Typical range for many biopharmaceuticals |
| Typical storage temperature | 2-8 °C | Some products require -20 °C or lower |
| Reconstitution time | Seconds to several minutes | Depends on cake porosity and diluent |
| Common moisture method | Karl Fischer titration | Measures water content in the solid |
| Container closure | Stoppered vial with seal | Protects against moisture and oxygen ingress |
Lyophilized products are typically stored as sealed solids in vials or syringes. Moisture ingress is a major concern because many dried cakes are hygroscopic and can lose stability when exposed to humid air. Storage temperature depends on the formulation; some products are kept refrigerated, while others are stable at room temperature. Container closure integrity and headspace moisture are often monitored. Light protection may also be required for some photosensitive materials.
Quality control for lyophilized materials includes visual inspection, residual moisture measurement, and reconstitution testing. Cake appearance can reveal process problems such as collapse, shrinkage, or meltback, although appearance alone does not prove potency. Residual moisture is commonly measured by Karl Fischer titration or by loss on drying. Reconstitution time is checked because a slow or incomplete dissolve can indicate a change in pore structure. Stability studies track these attributes over time under defined temperature and humidity conditions.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Regulatory expectations for lyophilized products focus on consistent manufacture and documented stability. Batches are often monitored for moisture, appearance, potency, and sterility where applicable. Process parameters such as shelf temperature, chamber pressure, and drying time are recorded and controlled within validated ranges. Open questions remain about how best to predict long-term stability from short accelerated studies, especially for complex biologics. Variations in freezing rate and ice crystal size can produce differences that are not always visible but may affect performance.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture uptake. Residual water content is measured because small changes can alter chemical stability and cake appearance. Storage temperature depends on the material; many biological products are kept at 2–8 °C, while some require −20 °C or colder. Exposure to ambient humidity during handling can cause the porous solid to absorb water and collapse. Container closures and stoppers are therefore selected for low moisture transmission and compatibility.
Quality control for lyophilized materials includes visual inspection of the cake, measurement of residual moisture, and tests for reconstitution time. An acceptable cake is typically uniform and may be slightly porous; shrinkage, meltback, or cracks can indicate process deviations. Analytical methods such as Karl Fischer titration, thermogravimetric analysis, and near-infrared spectroscopy quantify water content. Reconstitution time is recorded because a very slow or incomplete dissolution can signal collapse or aggregation. Stability studies compare samples stored under defined temperature and humidity conditions over months or years.
=== Protein tyrosine phosphatases === Protein Tyrosine Phosphatase (PTPs) are a group of enzymes that possess a catalytic domain with phosphotyrosine-specific phosphohydrolase activity. PTPs are capable of modifying the activity of receptor tyrosine kinases in both a positive and negative manner. PTPs can dephosphorylate the activated phosphorylated tyrosine residues on the RTKs which virtually leads to termination of the signal. Studies involving PTP1B, a widely known PTP involved in the regulation of the cell cycle and cytokine receptor signaling, has shown to dephosphorylate the epidermal growth factor receptor and the insulin receptor. Some PTPs, on the other hand, are cell surface receptors that play a positive role in cell signaling proliferation. Cd45, a cell surface glycoprotein, plays a critical role in antigen-stimulated dephosphorylation of specific phosphotyrosines that inhibit the Src pathway.

The carbon atom next to the carboxyl group is called the α–carbon. In proteinogenic amino acids, it bears the amine and the R group or side chain specific to each amino acid, as well as a hydrogen atom. With the exception of glycine, for which the side chain is also a hydrogen atom, the α–carbon is stereogenic. All chiral proteinogenic amino acids have the L configuration, and can therefore be referred to as L-amino acids. They are "left-handed" enantiomers, which refers to the stereoisomers of the alpha carbon. A few D-amino acids ("right-handed") have been found in nature, e.g., in bacterial envelopes, as a neuromodulator (D-serine), and in some antibiotics. Rarely, D-amino acid residues are found in proteins, and are converted from the L-amino acid as a post-translational modification.
=== Natural occurrence === Earth has retained all of the noble gases that were present at its formation except helium. Krypton's concentration in the atmosphere is about 1 ppm. It can be extracted from liquid air by fractional distillation. The amount of krypton in space is uncertain, because measurement is derived from meteoric activity and solar winds. The first measurements suggest an abundance of krypton in space.

On 15 October, when President Abdirashid Ali Sharmarke was touring Las Anood, his bodyguard killed him, he was acting on his own accord, according to Henry Kissinger. Six days later, on 21 October, General Siad Barre led a military coup which overthrew the parliamentary government. The coup was motivated by corruption. The bodyguard was tried and executed by the Supreme Revolutionary Council (SRC). He came from the same clan background as the president. Alongside Barre, the SRC was led by Brigadier General Mohamed Ainanshe Guled, Lieutenant Colonel Salaad Gabeyre Kediye, and Chief of Police Jama Korshel. Kediye officially held the title "Father of the Revolution", and afterwards, Barre became the head of the SRC. The SRC subsequently renamed the country to the Somali Democratic Republic, dissolved the parliament and the Supreme Court, and suspended the constitution.
Lake Afrera (in Italian Lake Giuletti) is a hypersaline lake in northern Ethiopia. Located in Kilbet Rasu, Afar Region, it is one of the lakes of the Danakil Depression. It has a surface of 117 km2 (45 mi2) and holds 2.4 km3 of brine. The maximal depth of Lake Afdera is 80 m in the northern basin and 76 m in the southern basin. No major river drains the lake. It is fed by hot springs along the shores of the lakes. It is also known as Lake Giulietti, the name Raimondo Franchetti gave it after the Italian explorer Giuseppe Maria Giulietti was slain by Afars southwest of the lake. Another name for this body of water is Lake Egogi (or Egogi Bad), which is the name L. M. Nesbitt's Afar guide gave it when the Italian explorer became the first European to see it in 1928. The single island in Lake Afrera, Franchetti Island (also known as "Deset"), located in the southern part of the lake, is considered the lowest-lying island in the world. Unlike other saline lakes in Ethiopia (e.g., Lakes Abijatta, Shala, and Chitu), the pH of Lake Afrera is low and in the acidic range. Although little studied, a few species of fish are hosted by Lake Afrera, including two endemics: Danakilia franchettii (a cichlid) and Aphaniops stiassnyae (syn. Lebias stiassnyae; a pupfish).
On April 12, 2001, the CPL Winter Championship commenced at the Hyatt Regency in Dallas, Texas. The event featured 48 competing teams, marking the first large-scale Counter-Strike tournament. The competition attracted over 800 attendees, including participants and Bring Your Own Computer (BYOC) players. Many professional Quake players were notably absent, while the Counter-Strike community saw an influx of new competitors. This tournament was the largest Counter-Strike event at the time and contributed to the game's rise in professional esports. In 2003, the Esports World Convention hosted a Counter-Strike tournament which featured 37 teams competing for a $100,000 prize pool. In 2012, the Electronic Sports League removed the game from the main titles for the 2012–2013 Intel Extreme Masters due to its decreasing popularity outside of Europe.

Unlike animals, many plant cells, particularly those of the parenchyma, do not terminally differentiate, remaining totipotent with the ability to give rise to a new individual plant. Exceptions include highly lignified cells, the sclerenchyma and xylem which are dead at maturity, and the phloem sieve tubes which lack nuclei. While plants use many of the same epigenetic mechanisms as animals, such as chromatin remodelling, an alternative hypothesis is that plants set their gene expression patterns using positional information from the environment and surrounding cells to determine their developmental fate. Epigenetic changes can lead to paramutations, which do not follow the Mendelian heritage rules. These epigenetic marks are carried from one generation to the next, with one allele inducing a change on the other.
==== Homeopathic HCG for weight control ==== Controversy about, and shortages of, injected HCG for weight loss have led to substantial Internet promotion of "homeopathic HCG" for weight control. The ingredients in these products are often obscure, but if prepared from true HCG via homeopathic dilution, they contain either no HCG at all or only trace amounts. Moreover, it is highly unlikely that oral HCG is bioavailable due to the fact that digestive protease enzymes and hepatic metabolism renders peptide-based molecules (such as insulin and human growth hormone) biologically inert. HCG can likely only enter the bloodstream through injection. The United States Food and Drug Administration has stated that over-the-counter products containing HCG are fraudulent and ineffective for weight loss. They are also not protected as homeopathic drugs and have been deemed illegal substances. HCG is classified as a prescription drug in the United States and it has not been approved for over-the-counter sales by the FDA as a weight loss product or for any other purposes, and therefore neither HCG in its pure form nor any preparations containing HCG may be sold legally in the country except by prescription. In December 2011, FDA and FTC started to take actions to pull unapproved HCG products from the market. In the aftermath, some suppliers started to switch to "hormone-free" versions of their weight loss products, where the hormone is replaced with an unproven mixture of free amino acids or where radionics is used to transfer the "energy" to the final product.
=== Prokaryotic origins === Ubiquitin is believed to have descended from bacterial proteins similar to ThiS (O32583) or MoaD (P30748). These prokaryotic proteins, despite having little sequence identity (ThiS has 14% identity to ubiquitin), share the same protein fold. These proteins also share sulfur chemistry with ubiquitin. MoaD, which is involved in molybdopterin biosynthesis, interacts with MoeB, which acts like an E1 ubiquitin-activating enzyme for MoaD, strengthening the link between these prokaryotic proteins and the ubiquitin system. A similar system exists for ThiS, with its E1-like enzyme ThiF. It is also believed that the Saccharomyces cerevisiae protein Urm1, a ubiquitin-related modifier, is a "molecular fossil" that connects the evolutionary relation with the prokaryotic ubiquitin-like molecules and ubiquitin. Archaea have a functionally closer homolog of the ubiquitin modification system, where "sampylation" with SAMPs (small archaeal modifier proteins) is performed. The sampylation system only uses E1 to guide proteins to the proteosome. Proteoarchaeota, which are related to the ancestor of eukaryotes, possess all of the E1, E2, and E3 enzymes plus a regulated Rpn11 system. Unlike SAMP which are more similar to ThiS or MoaD, Proteoarchaeota ubiquitin are most similar to eukaryotic homologs.
=== Expansion === In March 2020, many of the Cane's locations switched from dine-in to pick-up and take-out service only due to the COVID-19 pandemic, whereas others closed temporarily. As of July 2020, certain locations had reopened their dining rooms, although Graves said the company was in no rush to do so on a full scale. In 2021, in response to a shortage of workers at the time, the company began dispatching hundreds of its corporate employees to work in its restaurants as cooks and cashiers, in addition to their existing duties regarding the hiring of new employees. The company planned to hire 10,000 new employees. The company's co-CEO said that the corporate employees are trained in the kitchen and on the register under normal circumstances. In 2022, Raising Cane's sued a shopping center in Hobart, Indiana. After the restaurant chain had signed a long-term lease, it was revealed that the shopping center had a non-compete agreement with McDonald's which prohibited other vendors from selling de-boned chicken products in the complex. On June 27, 2023, Raising Cane's opened up their first New York location in Times Square.

Residual moisture can influence chemical degradation, cake collapse, and long-term stability. Low moisture levels usually improve stability, but each product has an optimal range.
Most lyophilized products are stored upright at controlled temperatures, often refrigerated or frozen. Protection from moisture and light helps maintain the dried cake.
A diluent is added to the dried cake, which dissolves to form a solution or suspension. Gentle mixing avoids foaming and preserves sensitive molecules.
Lyophilized products should be stored in airtight containers, protected from moisture and light, at the temperature specified by the manufacturer. Many require refrigeration at 2–8 °C, while some need frozen storage. Always check the product label for specific conditions.
