Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.
Storage stability depends on water content, oxygen exposure, and temperature. Lyophilized solids are hygroscopic and can absorb water if exposed to humid air. Vials are usually sealed under vacuum or inert gas with rubber stoppers and aluminum crimps. Storage temperatures range from room temperature to refrigerated or frozen conditions depending on the material. Stability studies track potency, moisture, and physical form over time. Accelerated conditions can reveal sensitivity but may not predict long-term behavior.
Quality assessment of a lyophilized product includes cake appearance, residual moisture, reconstitution time, and container closure integrity. A uniform, porous cake suggests that freezing and drying stayed within the formulation's design space. Cracks, shrinkage, meltback, or a glassy film can indicate thermal abuse or a formulation problem. Analysts also test for subvisible particles and sterility when the product requires those specifications. Visual inspection alone cannot confirm biological activity or chemical stability, so it is combined with analytical methods.
Stability programs monitor lyophilized products under defined temperature and humidity conditions over time. Real-time studies at recommended storage conditions are the reference, while accelerated studies provide early signals of degradation pathways. Because a dry cake can still undergo oxidation, hydrolysis, or aggregation, stability depends on residual moisture, excipients, and container headspace. Open questions include how best to predict long-term stability from short accelerated runs and how vial-to-vial variability affects shelf life. Current guidance treats these predictions as product-specific rather than universally generalizable.
| Property | Value | Notes |
|---|---|---|
| Appearance | White to off-white porous cake | Color and texture vary with formulation. |
| Reconstitution time | Seconds to several minutes | Depends on cake porosity, excipients, and diluent. |
| Typical moisture level | 0.5-3% w/w | Lower values suit hydrolysis-sensitive materials. |
| Common moisture method | Karl Fischer titration | Coulometric mode is common for low water levels. |
| Typical storage temperature | 2-8 °C or ambient | Some products require frozen storage; protect from humidity. |
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.
Stability of a lyophilized solid depends on water content, temperature, and the physical state of the formulation. Amorphous products may slowly absorb moisture and drop below their glass transition temperature, causing collapse or crystallization. Some proteins and peptides can aggregate even in a dry state, especially when exposed to heat or moisture. Accelerated stability studies at elevated temperature and humidity help estimate shelf life, but real-time data remain the basis for expiration dating.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
=== GnRH analogues === Analogs of gonadotropin-releasing hormone (GnRH) can be used to induce a chemical castration, that is, complete suppression of the production of estrogen and progesterone from the female ovaries, or complete suppression of testosterone production from the male testes. This is due to a negative feedback effect of continuous stimulation of the pituitary gland by these hormones. Leuprorelin and goserelin are GnRH analogs which are used primarily for the treatment of hormone-responsive prostate cancer. Because the initial endocrine response to GnRH analogs is actually hypersecretion of gonadal steroids, hormone receptor antagonists such as flutamide are typically used to prevent a transient boost in tumor growth.
With the arrival of Mia and Chloe, producers took the opportunity to explore Ari's fictional backstory further, which Kipa-Williams relished. He thought Ari and Mia were "star-crossed lovers" and believed that Ari had never stopped loving her. The pair soon reconcile, but their relationship is tested by his involvement with a criminal gang and a targeted hit-and-run, which leaves him with serious injuries. Writers scripted a pregnancy for the couple, however, Mia is involved in a car accident during the 2021 mid-season finale, and she has a miscarriage. At the end of the year, Chloe's biological father Matthew Montgomery (James Sweeny) was introduced and Ari takes the blame for his death when Chloe accidentally kills him while protecting Mia. Kipa-Williams quit Home and Away and producers decided to kill-off his character. Ari is admitted to hospital with cancer of the appendix and he opts not to have surgery to remove the tumours, as he wants to die on his terms. Ari and Mia marry shortly before he dies. He made his final appearance on 15 March 2022. Kipa-Williams received a nomination for Best Daytime Star at the Inside Soap Awards in 2020. Ari and the Parata family received a positive response from critics and viewers, with some branding them New Zealand's answer to the River Boys, a popular group of characters introduced in 2011. The character's break-up with Mac was not well received, with Digital Spy's Susannah Alexander and Daniel Kilkelly saying the show had "lost a solid couple".
=== Beyond Meat === In July 2018, A&W locations began serving Beyond Meat's vegan Beyond Burger. The chain had a shortage of the Beyond Burger in August 2018, but announced that all locations would receive stock by October 2018. In 2019 A&W expanded its Beyond Meat offerings with the release of the Beyond Meat Sausage N' Egger.
All other fats humans need can be made, at the cost of some energy, from those fats, or from other chemicals. Transfats are a class of fats which are very rare in nature, but very common in industrially processed oils as a consequence of "hydrogenation". It is becoming increasingly clear that ingestion of more than small quantities of transfats distorts some aspects of human biochemistry in ways which increase artery disease and heart disease, and so increase death rates. Saturated fats have a much smaller deleterious health effect. Fatty acids chains of carbon atoms with attached side groups. They are found in living tissues, typically in the form of triglycerides (three fatty acid chains attached to a glycerol backbone). Fiber usually carbohydrate which cannot be digested. It passes through the human digestive system without being digested or absorbed. Soluble fiber absorbs water in the intestines, insoluble does so much less. Fiber has effects on intestinal operations, and by extension, on other tissues. For instance, adequate fiber intake seems to have an effect on vascular health. The mechanisms by which these effects happen are largely speculative at present. One effect of dietary fiber, apparently especially soluble fiber, has is to increase stool size and softness. Dietary fibers, when eaten together with carbohydrates, delay the uptake of the carbohydrates. ==== Paid leave while on Family and Medical Leave Act leave ==== The temporary tax credit for employers who give paid leave to an employee while they are on Family and Medical Leave Act leave has been made permanent. In order to qualify for the tax credit, the employer must pay the employee at least 50% of the employee's usual wages. The amount of the tax credit used to be equal to 12.5% of eligible wages paid to an eligible employee, but it now increases by 0.25% for each percentage point paid above the 50% threshold, up to a maximum credit of 25%. In order to be eligible for the tax credit, the employee must have worked at least six months for the employer, which is up from 12 months previously. Paid family and medical leave that is required under state or local law is now eligible for the tax credit, as are employer-paid amounts for qualifying paid leave insurance policies.
=== Businesses === In October 2020, the e-commerce platform Shopify added TikTok to its portfolio of social media platforms, allowing online merchants to sell their products directly to consumers on TikTok. Some small businesses have used TikTok to advertise and to reach an audience wider than the geographical region they would normally serve. The viral response to many small business TikTok videos has been attributed to TikTok's algorithm, which shows content that viewers at large are drawn to, but which they are unlikely to actively search for (such as videos on unconventional types of businesses, like beekeeping and logging). In 2020, digital media companies such as Group Nine Media and Global used TikTok increasingly, focusing on tactics such as brokering partnerships with TikTok influencers and developing branded content campaigns. Notable collaborations between larger brands and top TikTok influencers have included Chipotle's partnership with David Dobrik in May 2019 and Dunkin' Donuts' partnership with Charli D'Amelio in September 2020.

Karl Fischer titration is a common method, using coulometric or volumetric detection. Thermogravimetric analysis can also measure weight loss on heating. Results depend on sample handling because the dried solid can absorb moisture quickly.
The porous cake readily absorbs water vapor from air, which can reduce stability or cause collapse. Vials are sealed with stoppers and crimp seals, sometimes under vacuum or inert gas. Packaging also protects against oxygen and mechanical damage.
Collapse occurs when the product temperature rises above its collapse threshold during primary drying. The ice matrix loses structure, and the cake may shrink or melt back. Formulation excipients and freezing rate influence collapse threshold.
Many dried cakes are hygroscopic and can adsorb water during storage or handling. Absorbed moisture may lower the glass transition temperature and promote chemical reactions. Sealed packaging and controlled humidity reduce this risk.
