This is a working overview of Eutectic point, written for readers who want more than a one-paragraph summary but less than a textbook.

Lyophilized products are typically stored as sealed solids in vials or syringes. Moisture ingress is a major concern because many dried cakes are hygroscopic and can lose stability when exposed to humid air. Storage temperature depends on the formulation; some products are kept refrigerated, while others are stable at room temperature. Container closure integrity and headspace moisture are often monitored. Light protection may also be required for some photosensitive materials.
Quality control for lyophilized materials includes visual inspection, residual moisture measurement, and reconstitution testing. Cake appearance can reveal process problems such as collapse, shrinkage, or meltback, although appearance alone does not prove potency. Residual moisture is commonly measured by Karl Fischer titration or by loss on drying. Reconstitution time is checked because a slow or incomplete dissolve can indicate a change in pore structure. Stability studies track these attributes over time under defined temperature and humidity conditions.
Analytical methods for lyophilized solids must account for the low moisture content and the fragile cake. Karl Fischer titration is widely used for water content, while near-infrared spectroscopy can measure moisture non-destructively in sealed containers. X-ray diffraction and modulated differential scanning calorimetry help identify crystalline or amorphous phases. Residual solvent analysis may be needed if organic solvents were used during formulation. The combination of these methods supports batch release and long-term stability assessment.
The physics of lyophilization depends on phase boundaries and heat and mass transfer. During primary drying, heat supplied to the product must equal the latent heat of sublimation, while water vapor moves through the drying cake to the condenser. If shelf temperature or pressure is set too high, the ice front can exceed the collapse temperature, causing meltback or pore collapse. If conditions are too cold, drying slows and costs rise. Formulation excipients, vial geometry, and freezing rate alter these limits.
Equipment for lyophilization includes a vacuum chamber, temperature-controlled shelves, a condenser, and a vacuum pump. A refrigeration system cools the shelves and condenser below the product's freezing point. Process monitoring often uses Pirani and capacitance manometers, thermocouples, and resistance sensors. Cycle development balances product quality with time and energy use. Some products are annealed during freezing to improve crystallization of bulking agents. Open questions remain about scaling cycles between laboratory, pilot, and production freeze-dryers.
Lyophilization, also called freeze-drying, removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts free water into ice and may also produce a glassy phase. Primary drying then lowers chamber pressure so ice sublimes directly to vapor without passing through a liquid stage. Secondary drying raises the temperature modestly to remove bound water. The result is a porous, dry solid that usually reconstitutes quickly. Each stage influences pore structure, residual moisture, and stability.
| Property | Value | Notes |
|---|---|---|
| Storage temperature | 2–8 °C or 20–25 °C | Depends on product stability; some require frozen storage. |
| Moisture content | 0.5–3% w/w | Higher values may reduce stability; target set per product. |
| Moisture method | Karl Fischer titration | Coulometric for low levels; volumetric for higher levels. |
| Cake appearance | Uniform, intact, no collapse | Visual inspection is qualitative and not a potency measure. |
| Reconstitution time | Seconds to several minutes | Depends on cake density, excipients, and diluent. |
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
The process usually has three stages: freezing, primary drying, and secondary drying. Freezing sets the ice crystal structure and can determine pore size in the final cake. Primary drying removes free ice by sublimation at low pressure and low shelf temperature. Secondary drying removes bound water by raising the shelf temperature, although some water may remain as part of the solid. Cycle parameters depend on formulation, fill volume, vial type, and equipment performance.
The physical chemistry of freezing influences whether a formulation forms an amorphous glass or a crystalline solid. Amorphous systems can collapse if product temperature rises above the glass transition temperature of the freeze concentrate. Crystalline systems may show eutectic melting, where ice and solute melt together at a fixed temperature. Formulators add bulking agents, lyoprotectants, and buffers to preserve structure and biological activity. The optimum cycle keeps product temperature below critical thresholds during primary drying while allowing efficient sublimation.
Lyophilization is a dehydration process that removes water from a frozen material by sublimation under low pressure. The material is first frozen so that water becomes ice; then the surrounding pressure is lowered below the vapor pressure of ice. Heat is applied gently so ice changes directly to vapor without passing through a bulk liquid phase. The vapor is collected on a cold condenser, leaving a dry porous matrix. This process differs from simple evaporation because the material remains frozen during the main drying stage.
Similarly, altered somatosensory integration and introspection may relate to abnormal body image. A review of functional neuroimaging studies reported reduced activations in "bottom up" limbic region and increased activations in "top down" cortical regions which may play a role in restrictive eating. Compared to controls, people who have recovered from anorexia show reduced activation in the reward system in response to food, and reduced correlation between self reported liking of a sugary drink and activity in the striatum and anterior cingulate cortex. Increased binding potential of 11C radiolabelled raclopride in the striatum, interpreted as reflecting decreased endogenous dopamine due to competitive displacement, has also been observed. Structural neuroimaging studies have found global reductions in both gray matter and white matter, as well as increased cerebrospinal fluid volumes. Regional decreases in the left hypothalamus, left inferior parietal lobe, right lentiform nucleus and right caudate have also been reported in acutely ill patients. However, these alterations seem to be associated with acute malnutrition and largely reversible with weight restoration, at least in nonchronic cases in younger people. In contrast, some studies have reported increased orbitofrontal cortex volume in currently ill and in recovered patients, although findings are inconsistent. Reduced white matter integrity in the fornix has also been reported.
In 1928, it became a separate province. Between 1914 and 1928, the Ma clique ruled the provinces of Qinghai, Ningxia and Gansu; General Ma Hongkui was the military governor of Ningxia and had absolute authority in the province. The Muslim conflict in Gansu, which lasted from 1927 to 1930, spilt over into Ningxia. In 1934, warlord and National Revolutionary Army general Sun Dianying attempted to conquer the province, but was defeated by an alliance led by the Ma clique. From 1950 to 1958, a Kuomintang Islamic insurgency resulted in fighting throughout Northwest China, including Ningxia. In 1954, the Chinese government merged Ningxia with Gansu, but in 1958 Ningxia formally became an autonomous region of China. In 1969, Ningxia received a part of the Inner Mongolian Autonomous Region, but this area was returned in 1979. A number of Chinese artifacts dating from the Tang dynasty and Song dynasty, some of which had been owned by Emperor Zhenzong, were excavated and then came into the hands of Ma Hongkui, who refused to publicize the findings. Among the artifacts were a white marble tablet from the Tang dynasty, gold nails, and bands made out of metal. It was not until after Ma Hongkui died that his wife went to Taiwan in 1971 from America to bring the artifacts to Chiang Kai-shek, who turned them over to the Taipei National Palace Museum.
== Characterization == Volatile thiols are easily and almost unerringly detected by their distinctive odor. Sulfur-specific analyzers for gas chromatographs are useful. Spectroscopic indicators are the D2O-exchangeable SH signal in the 1H NMR spectrum (33S is NMR-active but signals for divalent sulfur are very broad and of little utility). The νSH band appears near 2400 cm−1 in the IR spectrum. In the nitroprusside reaction, free thiol groups react with sodium nitroprusside and ammonium hydroxide to give a red colour.
=== Structural motifs === In addition to linear degrons, the E3 ligase can in some cases also recognize structural motifs on the substrate. In this case, the 3D motif can allow the substrate to directly relate its biochemical function to ubiquitination. This relation can be demonstrated with TRF1 protein (regulator of human telomere length), which is recognized by its corresponding E3 ligase (FBXO4) via an intermolecular beta sheet interaction. TRF1 cannot be ubiquinated while telomere bound, likely because the same TRF1 domain that binds to its E3 ligase also binds to telomeres.

=== Background === Banki's family lived in Iran and through the early 2000s with his parents' marriage falling apart, the family decided to send a portion of the family assets to the US. Between 2006 and 2009 Banki's family sent proceeds of his parents' divorce settlement (approximately $3.4 million) to him, from Iran to the US. The money came into a single bank account at Bank of America, over multiple transfers through an informal money transfer systems which the defendant argued was legal and the only means of sending money out of Iran at the time. Banki used the majority of this money to purchase an apartment in downtown Manhattan. Prosecutors argued that over this three-year period Banki had violated the sanctions imposed by the US on Iran. The indictment accused Banki of violating the sanctions law, running an informal underground bank, and helping "manage, supervise, operate, and conduct" an unlicensed money transmittal service through which he directly or indirectly facilitated and violated the Iran sanctions. In essence, the charges against Banki stemmed from prosecution allegations that he, as an American citizen, had violated US sanctions on Iran.
=== Other factors === Not getting enough sleep is also associated with obesity. Whether one causes the other is unclear. Even if short sleep does increase weight gain, it is unclear if this is to a meaningful degree or if increasing sleep would be of benefit. Some researchers have proposed that chemical compounds called "obesogens" (a category that includes certain endocrine-disrupting chemicals such as bisphenols and phthalates) may play a role in obesity. Certain aspects of personality are associated with being obese. Loneliness, neuroticism, impulsivity, and sensitivity to reward are more common in people who are obese while conscientiousness and self-control are less common in people who are obese. Because most of the studies on this topic are questionnaire-based, it is possible that these findings overestimate the relationships between personality and obesity: people who are obese might be aware of the social stigma of obesity and their questionnaire responses might be biased accordingly. Similarly, the personalities of people who are obese as children might be influenced by obesity stigma, rather than these personality factors acting as risk factors for obesity. In relation to globalization, it is known that trade liberalization is linked to obesity; research, based on data from 175 countries during 1975–2016, showed that obesity prevalence was positively correlated with trade openness, and the correlation was stronger in developing countries.
The reasoning was regarded by a majority of commentators as thinly veiled attempt of the Court of Justice to clutch onto its own power, but it has meant the commission is redrafting a new accession agreement. Under TEU articles 3(5), 21, 34 and 42, the EU must also respect the principles of the United Nations Charter. After the September 11 attacks on the World Trade Center in New York City, the UN Security Council adopted a resolution to freeze the assets of suspected terrorists, linked to Osama bin Laden. This included a Saudi national, Mr Kadi. Sweden froze his assets pursuant to an EU Regulation, which gave effect to the UN Security Council resolution. In Kadi v Commission, Mr Kadi claimed there was no evidence that he was connected to terrorism, and he had not had a fair trial: a fundamental human right. The opinion of AG Maduro recalled Aharon Barak, of the Supreme Court of Israel, that it "is when the cannons roar that we especially need the laws". The Court of Justice held that even UN member cannot contravene "the principles that form part of the very community legal order". In effect the EU has developed a rule that within the boundaries of certain jus cogens principles, other courts may take primacy. The content of those core principles remains open to ongoing judicial dialogue among the senior courts in the Union.

In overt primary hyperthyroidism, TSH levels are low, and T4 and T3 levels are high. Subclinical hyperthyroidism is a milder form of hyperthyroidism characterized by low or undetectable serum TSH level, but with a normal serum free thyroxine level. Although the evidence for doing so is not definitive, treatment of elderly persons having subclinical hyperthyroidism could reduce the number of cases of atrial fibrillation. There is also an increased risk of bone fractures (by 42%) in people with subclinical hyperthyroidism; there is insufficient evidence to say whether treatment with antithyroid medications would reduce that risk. A 2022 meta-analysis found subclinical hyperthyroidism to be associated with cardiovascular death.
However, it was soon realized that submarines could approach enemy coastlines undetected and decrease the warning time (the time between detection of the missile launch and the impact of the missile) from as much as half an hour to possibly under three minutes. This effect was especially significant to the United States, Britain and China, whose capitals of Washington D.C., London, and Beijing all lay within 100 miles (160 km) of their coasts. Moscow was much more secure from this type of threat, due to its considerable distance from the sea. This greatly increased the credibility of a "surprise first strike" by one faction and (theoretically) made it possible to knock out or disrupt the chain of command of a target nation before any counterstrike could be ordered (known as a "decapitation strike"). It strengthened the notion that a nuclear war could possibly be "won", resulting not only in greatly increased tensions and increasing calls for fail-deadly control systems, but also in a dramatic increase in military spending. The submarines and their missile systems were very expensive, and one fully equipped nuclear-powered and nuclear-armed missile submarine could cost more than the entire GNP of a developing country. It was also calculated, however, that the greatest cost came in the development of both sea- and land-based anti-submarine defenses and in improving and strengthening the "chain of command", and as a result, military spending skyrocketed. South Africa developed a nuclear weapon capability during the 1970s and early 1980s.
=== Electrophysiology === Electrophysiology experiments were used in the early days of neuroendocrinology to identify the physiological happenings in the hypothalamus and the posterior pituitary especially. In 1950, Geoffrey Harris and Barry Cross outlined the oxytocin pathway by studying oxytocin release in response to electrical stimulation. In 1974, Walters and Hatton investigated the effect of water dehydration by electrically stimulating the supraoptic nucleus—the hypothalamic center responsible for the release of vasopressin. Glenn Hatton dedicated his career to studying the physiology of the Neurohypophyseal system, which involved studying the electrical properties of hypothalamic neurons. Doing so enabled investigation into the behavior of these neurons and the resulting physiological effects. Studying the electrical activity of neuroendocrine cells enabled the eventual distinction between central nervous neurons, neuroendocrine neurons, and endocrine cells.
The MEN-1 syndrome often exhibits tumors of parathyroid glands, anterior pituitary, endocrine pancreas, and endocrine duodenum. Less frequently, neuroendocrine tumors of lung, thymus, and stomach or non-endocrine tumors such as lipomas, angiofibromas, and ependymomas are observed neoplasms. In a study of 12 sporadic carcinoid tumors of the lung, five cases involved inactivation of both copies of the MEN1 gene. Of the five carcinoids, three were atypical and two were typical. The two typical carcinoids were characterized by a rapid proliferative rate with a higher mitotic index and stronger Ki67 positivity than the other typical carcinoids in the study. Consequently, the carcinoid tumors with MEN1 gene inactivation in the study were considered to be characterized by more aggressive molecular and histopathological features than those without MEN1 gene alterations.

Most lyophilized products are stored in sealed containers at controlled temperature and humidity. Some require refrigeration, while others are stable at room temperature. Protection from light and moisture is often necessary.
Cake collapse indicates that the porous structure was lost during drying. It can result from excessive product temperature or an unsuitable formulation. Collapsed cakes may have slower reconstitution and are often rejected by visual inspection.
Residual moisture affects the chemical and physical stability of a lyophilized solid. High moisture can promote degradation, aggregation, or cake shrinkage. The acceptable range is set for each product based on stability data.
Simple drying usually removes water by evaporation from a liquid or solid, often with heat. Lyophilization first freezes the material and then removes ice by sublimation under vacuum. This avoids prolonged exposure to liquid water and high temperatures.
